APPdup cells secrete more Aβ40 and Aβ42 peptides than controls
Endothelial cells from Down syndrome and APPdup both secrete elevated levels of Aβ40 and Aβ42 compared to controls, yet they exhibit divergent barrier integrity. Cells from Down syndrome show increased junctional protein expression and reduced permeability, while APPdup cells display increased permeability.
- Field
- the brain and neuroscience
- What they did
- Researchers investigated endothelial cell function and blood-brain barrier integrity in individuals with Down syndrome and APP duplication using stem cell-derived cells. They found that while cells in Down syndrome produce more amyloid-beta, they have tighter junctions and lower permeability, whereas cells in APP duplication show increased permeability.
- Why it matters
- This difference in cell behavior helps explain why amyloid deposition in the brain's blood vessels occurs more rapidly and severely in APP duplication compared to Down syndrome, despite both conditions involving an extra copy of chromosome 21.
